Existing banked and well-characterized tissue collections are a treasure chest of genetic information; however, DNA and RNA from FFPE tissues is highly degraded. AltheaDx’s methods can extract highly valuable information from these samples. AltheaDx provides a range of Short Fragment Assay (SFA) formats for the analysis of degraded samples, including FFPE tissue blocks. We have extensive experience in high-throughput sample analysis using our propriety XP™- PCR technology to monitor gene expression in highly degraded RNA samples. Sample AltheaDx Results: Multiplexed XP-PCR from FFPE Tissues
RNA samples were derived from fresh frozen prostate tumor tissue and from a 16 year-old FFPE tissue block. Ten reference genes were quantified using single multiplex XP-PCR reactions and real-time PCR. Even in the degraded fixed tissue sample (top right), XP-PCR generated good signal outperforming real-time PCR (lower right). |
