REAL-TIME ASSAYS USING TAQMAN, LIGHTCYCLER AND SCORPIONS CHEMISTRIES

AltheaDx principally runs real-time assays using TaqMan and DxS Scorpions chemistries. Experimental CVs average less than 5% using triplicate data in gene expression studies. Assays sensitivities can run down to a few percent in genotyping studies. Designing quality, specific assays using TaqMan, Scorpions or Scorpions plus ARMS offers real challenges and the AltheaDx scientific staff has extensive experience in primer and probe design and testing. All assays undergo full qualification in house, with specific assays undergoing complete validation as required by the customer and the FDA. Assays tracking towards IVD can be developed with integrated design control under QSR.

All RNA analysis is designed to be performed with the reverse transcription step in multiplex format and the PCR in either singlet or duplex format, one gene plus one reference gene. Single step RT-PCR designs can also be created. DNA analyses are typically performed in singlet or duplex format for biallelic SNP genotyping or wild type/variant mutation testing. Genotype discrimination can be performed either at the probe for TaqMan or the primer for Scorpions plus ARMS. AltheaDx currently designs for open tube format or AB TLDA format.

AltheaDx’s facilities include AB 7700 real-time instruments for the analysis of mid to large volume samples in 96-well format and 2 Roche Lightcycler 480II, 3 AB 7900HT real-time instruments and one AB/Life Tech ViiA™ 7 Real-Time PCR System with automation for the analysis of small volume samples in 96 or 384-well format, including the use of the AB TaqMan low density array (TLDA), microfluidic cards. AltheaDx has the capacity to process hundreds of plates of real-time PCR reactions each month under GLP.
