AltheaDx offers mutation scanning services by High Resolution Melting (HRM) using either Roche LightCycler or AB/Life Technologies ViiA™ 7 Real-Time PCR System instruments. HRM analysis is a post-PCR analysis method used to identify variations in nucleic acid sequences. HRM-PCR is a powerful technique for the detection of mutations, polymorphisms and epigenetic differences in double stranded DNA samples. The method is based on detecting small differences in PCR melting (dissociation) curves. It is enabled by applying optimized dsDNA-binding dyes in conjunction with real-time PCR instrumentation that has precise temperature ramp control and advanced data capture capabilities. Compared to other technologies for mutation scanning, such as pyrosequencing, denaturing HPLC (dHPLC), and denaturing gradient gel electrophoresis (DGGE), HRM offers cost and time efficiency, tight reproducibility, and a high level of accuracy and sensitivity. The turnaround time for HRM-PCR data delivery is only two to three business days (depending on the number of samples).
