EGFR Pathway
Biomarkers for the EGF Receptor Pathway
- Patient response to drugs targeting the Epidermal Growth Factor Receptor (EGFR) is dependent on mutations in the EGFR itself as well as its downstream signaling proteins
- Somatic mutations in the EGFR correlate with drug responsiveness
- Mutations in downstream signaling proteins can activate proliferation and inhibit drug response:
| - EGFR |
- B-Raf |
- P-Ten |
| - K-Ras |
- PI3K/AKT |
- Mek |
- Drugs targeting other members of the EGFR pathway are also affected by these mutations
- Understanding patient EGFR pathway mutation profiles may enable selection of more effective therapies
AltheaDx has capabilities for detecting mutations in genes known to be involved in multiple disease and drug-targeted signaling pathways:
- Point mutations (e.g. K-Ras, b-Raf) and SNP profiles are identified using real-time PCR methods
- Genes with broader mutation spectra (e.g. PI3K, Akt) are examined using targeted double-strand sequencing
AltheaDx continues to expand its repertoire of standard mutation detection tests, and seeks collaborations to identify new gene mutations and their linked pharmacology.
