EGFR Pathway

Biomarkers for the EGF Receptor Pathway

  • Patient response to drugs targeting the Epidermal Growth Factor Receptor (EGFR) is dependent on mutations in the EGFR itself as well as its downstream signaling proteins
  • Somatic mutations in the EGFR correlate with drug responsiveness
  • Mutations in downstream signaling proteins can activate proliferation and inhibit drug response:
- EGFR - B-Raf - P-Ten
 - K-Ras - PI3K/AKT - Mek
  • Drugs targeting other members of the EGFR pathway are also affected by these mutations
  • Understanding patient EGFR pathway mutation profiles may enable selection of more effective therapies

AltheaDx has capabilities for detecting mutations in genes known to be involved in multiple disease and drug-targeted signaling pathways:

  • Point mutations (e.g. K-Ras, b-Raf) and SNP profiles are identified using real-time PCR methods
  • Genes with broader mutation spectra (e.g. PI3K, Akt) are examined using targeted double-strand sequencing
